Model Description
Validation Data

Fig.1 Detection of MEFV expression in bone marrow, spleen and lung by RT-PCR. Human MEFV mRNA (421 bp) was detectable only in heterozygous Mefv-hMEFV*V726A knockin mice but not in wild-type mice. Bone marrow, spleen and lung RNA was extracted from 4-week-old male wild-type C57BL/6 (WT)(n=3) and heterozygous Mefv-hMEFV*V726A knockin mice (HE) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.
Abbr. M, marker; HE, heterozygous; WT, wild type.

Fig.2 mRNA sequencing of Mefv-hMEFV*V726A mice. Sequencing data of RT-PCR products confirmed the presence of target mutations in Mefv-hMEFV*V726A mice.
Abbr. HE, heterozygous; WT, wild type.

Fig.3 Detection of MEFV expression in Mefv-hMEFV*V726A mice by WB. MEFV was detectable in spleen, bone marrow and lung from both WT C57BL/6 and heterozygous Mefv-hMEFV*V726A mice, as the antibody was cross-reactived between human and mouse MEFV. The spleen, bone marrow and lung tissue lysates were collected from 4-week-old male wild-type C57BL/6 mice and 4-week-old male heterozygous Mefv-hMEFV*V726A mice, and then analyzed by western blot with anti-Mefv antibody.
Abbr. HE, heterozygous; WT, wild type.
