Fig.1 Detection of App expression in brain by RT-PCR. Brain RNA was extracted from wild-type C57BL/6 mice (WT, n=3, male, 11-12 weeks old) and homozygous hAppNL-F knockin mice (HO, n=3, male, 17 weeks old), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Target mRNA products were amplified using mouse-specific App primers (127 bp) and human-specific APP primers (516 bp) in both WT C57BL/6 and homozygous hAppNL-F knockin mice.

Fig.2 mRNA sequencing of hAppNL-F knockin mice. Sequencing data of RT-PCR products confirmed the presence of target mutations, ensuring the successful integration of the NL and F substitutions into the humanized APP sequence of hAppNL-F mice.

Fig.3 Detection of  APP expression in hAppNL-F knockin mice by WB. The whole brain tissues were collected from wild-type C57BL/6J mice (WT, male, 11-12 weeks old) and homozygous hAppNL-F knockin mice (HO, male, 17 weeks old), and then analyzed by western blot with anti-APP antibody (Abcam, ab32136). 20 μg of total proteins were loaded for western blotting analysis. The antibody is cross-reactive between human and mouse APP.
