Validation Data

Fig.1 Detection of Il5 expression in thymus by RT-PCR. Mouse Gapdh mRNA (123 bp) was detectable both in wild-type C57BL/6 mice and homozygous Il5-KO mice. Mouse Il5 mRNA (177 bp/175 bp) was detectable only in wild-type mice but not in homozygous Il5-KO mice. Thymus RNA was extracted from 5-week-old male wild-type C57BL/6 (WT) (n=3) and homozygous Il5-KO mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of mouse IL5 expression in serum of WT C57BL/6 mice and HO Il5-KO mice by ELISA. Mouse IL5 was exclusively detectable in wild-type C57BL/6 mice. Serum were collected from wild-type C57BL/6 mice (n=3, male, 5 weeks old) and HO Il5-KO mice (n=3, male, 5 weeks old), and analyzed by ELISA.

Abbr.  HO, homozygous; WT, wild type.
