Fig.2 Detection of NR1I2 expression in duodenum and large Intestine by RT-PCR. Mouse Nr1i2 mRNA (217 bp) was detectable only in wild-type C57BL/6 mice. Human NR1I2 mRNA (361 bp) was detectable only in homozygous hNR1I2 knockin mice but not in wild-type mice. Duodenum and large Intestine RNA was extracted from 6-week-old female wild-type C57BL/6 (WT)(n=2) and homozygous hNR1I2 knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Fig.3 Detection of NR1I2 expression in hNR1I2 mice by WB. NR1I2 was detectable in liver and colon from both WT C57BL/6 and HO hNR1I2 mice, as the antibody was cross-reactived between human and mouse NR1I2. The liver and colon tissue lysates were collected from 7-week-old female wild-type C57BL/6 mice and 6-week-old female homozygous hNR1I2 mice, and then analyzed by western blot.
