Gene Summary

Gene Symbol
Lrp2

Model Description

The endogenous mouse Lrp2 gene was replaced by human LRP2 gene.

Validation Data

Fig.1 Detection of LRP2 expression in kidney by RT-PCR. Mouse Lrp2 mRNA (148 bp) was detectable only in wild-type C57BL/6 mice. Human LRP2 mRNA (124 bp) was detectable only in homozygous hLRP2 knockin mice but not in wild-type mice. Kidney RNA was extracted from 5-week-old male wild-type C57BL/6 (n=2) and homozygous hLRP2 knockin mice (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of LRP2 mRNA expression in kidney by qPCR. Mouse Lrp2 mRNA was detectable only in wild-type C57BL/6 mice. Human LRP2 mRNA was detectable only in homozygous hLRP2 knockin mice but not in wild-type mice. Kidney RNA was extracted from 5-week-old male wild-type C57BL/6 (n=2) and homozygous hLRP2 knockin mice (n=2), then cDNA libraries were synthesized by reverse transcription, followed by qPCR (A) with human LRP2 and mouse Gapdh primers and (B) with mouse Lrp2 and mouse Gapdh primers. Relative expression represents (A) the human LRP2 mRNA level relative to its average expression in homozygous hLRP2 knockin mice or (B) the mouse Lrp2 mRNA level relative to its average expression in wild-type mice.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.3 Detection of human LRP2 expression in hLRP2 knockin mice by IHC. Human LRP2 was exclusively detectable in hLRP2 knockin mice but not in WT mice. Human LRP2 was predominantly located on the brush border of kidney proximal convoluted tubules. Tissues were collected from 5-week-old male WT C57BL/6 mice and HO hLRP2 knockin mice, then analyzed by IHC with anti-human LRP2 antibody. Black arrows indicate positive staining (brown). The scales are same in each image and are 0.1mm. Magnification, 40X.

Abbr. HO, homozygous; WT, wild type.
