Alkbh5

Model Description

The endogenous mouse Alkbh5 gene was replaced by human ALKBH5 gene.

Validation Data

Fig.1 Detection of ALKBH5 expression in kidney by RT-PCR. Kidney RNA was extracted from 7-week-old male wild-type C57BL/6 (WT) and homozygous hALKBH5(2) knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Alkbh5 mRNA (621 bp) was detectable only in wild-type C57BL/6 mice. Human ALKBH5 mRNA (500 bp) was detectable only in homozygous hALKBH5(2) knockin mice but not in wild-type mice.

Fig.2 Detection of ALKBH5 expression in hALKBH5(2) mice by WB. The lysates of brain, skeletal muscle and eWAT tissues were collected from 7-week-old male WT C57BL/6 mice and homozygous hALKBH5(2) mice, and then analyzed by western blot with anti-ALKBH5 antibody. ALKBH5 was detectable in brain, skeletal muscle and eWAT tissues from both WT C57BL/6 and HO hALKBH5(2) mice, as the antibody was cross-reactived between human and mouse ALKBH5.

Abbr. HO, homozygous; WT, wild type; PC, positive control, Hela cells; M, marker; eWAT, epididymal white adipose tissue.
