Gene Summary

Gene Symbol
Ager

Model Description

The endogenous mouse Ager gene was replaced by human AGER gene.

Validation Data

Fig.1 Detection of AGER expression in lung by RT-PCR. Mouse Ager mRNA (204 bp) was detectable only in wild-type C57BL/6 mice. Human AGER mRNA (169 bp) was detectable only in homozygous hAGER knockin mice but not in wild-type mice. Lung RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=3) and homozygous hAGER knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of human AGER expression in serum of WT C57BL/6 mice and HO hAGER mice by ELISA. Human AGER was exclusively detectable in homozygous hAGER knockin mice. Serum were collected from wild-type C57BL/6 mice (n=3, male, 6 weeks old) and HO hAGER mice (n=3, male, 6 weeks old), and analyzed by ELISA with anti-hRAGE ELISA kit.

Abbr.  HO, homozygous; WT, wild type; N.D, not detected.

Fig.3 Detection of human AGER expression in lung homogenate of WT C57BL/6 mice and HO hAGER mice by ELISA. Human AGER was exclusively detectable in homozygous hAGER knockin mice. Lung homogenate were collected from wild-type C57BL/6 mice (n=3, male, 6 weeks old) and HO hAGER mice (n=3, male, 6 weeks old), and analyzed by ELISA with anti-hRAGE ELISA kit.

Abbr.  HO, homozygous; WT, wild type; N.D, not detected.

Fig.4 Detection of AGER expression in hAGER mice by WB. Human AGER was exclusively detectable in lung from HO hAGER mice with a human AGER antibody. The lung tissue lysates were collected from 6-week-old male wild-type C57BL/6 mice and 6-week-old male homozygous hAGER mice, and then analyzed by western blot with anti-AGER antibody.

Abbr. HO, homozygous; WT, wild type.
