Fig.1 Detection of CD8 expression on T cells in blood and spleen of WT C57BL/6 mice and hCD19/hCD8A mice by FACS. Blood and spleen were collected from WT C57BL/6 mice (male, 11 weeks old) and homozygous hCD19/hCD8A humanized mice (male, 11 weeks old). CD8 expression was analyzed from CD3+ population. hCD8 was exclusively detectable in the blood and spleen of HO hCD19/hCD8A humanized mice but not in WT mice, whereas mCD8 was exclusively detectable on T cells in WT C57BL/6 mice.
Abbr. HO, homozygous; WT, wild type.

Fig.2 Detection of CD19 expression on B cells in blood and spleen of WT C57BL/6 mice and hCD19/hCD8A mice by FACS. Blood and spleen were collected from WT C57BL/6 mice (male, 11 weeks old) and homozygous hCD19/hCD8A humanized mice (male, 11 weeks old). CD19 expression was analyzed from CD3- population. hCD19 was exclusively detectable in the blood and spleen of HO hCD19/hCD8A humanized mice but not in WT mice, whereas mCD19 was exclusively detectable on B cells in WT C57BL/6 mice.
Abbr. HO, homozygous; WT, wild type.

Fig.3 Representative figure of leukocyte and myeloid subpopulations in blood in HO hCD19/hCD8A mice. T cells, B cells, NK cells, CD11b+myeloid cells, DCs, neutrophils, monocytes, and macrophages in the blood of HO hCD19/hCD8A mice showed comparable to those in WT C57BL/6 mice. Percentages of T cells, B cells, NK cells, CD11b+myeloid cells, DCs, neutrophils, monocytes, and macrophages in the blood of HO hCD19/hCD8A mice showed comparable to those in WT C57BL/6 mice.

Fig.4 Representative figure of leukocyte and myeloid subpopulations in spleen in HO hCD19/hCD8A mice. T cells, B cells, NK cells, CD11b+myeloid cells, DCs, neutrophils, monocytes, and macrophages in the spleen of HO hCD19/hCD8A mice showed comparable to those in WT C57BL/6 mice. Percentages of T cells, B cells, NK cells, CD11b+myeloid cells, DCs, neutrophils, monocytes, and macrophages in the spleen of HO hCD19/hCD8A mice showed comparable to those in WT C57BL/6 mice.

Fig.5 Frequency of leukocyte and myeloid subpopulations in blood and spleen by flow cytometry. Blood and spleen was collected from WT C57BL/6 mice and HO hCD19/hCD8A mice (10-week-old, n=3). Flow cytometry analysis of frequencies of leukocyte and myeloid subpopulations in (A) blood and (B) spleen. Bar indicated expressed as mean ± SEM. Percentages of T cells, B cells, NK cells,CD11b+myeloid cells, DCs, neutrophils, monocytes, and macrophages in the blood and spleen of HO hCD19/hCD8A mice showed comparable to those in WT C57BL/6 mice.
Abbr. HO, homozygous; WT, wild type.
