Fig.1 Detection of OX40 expression on Treg, CD4+T and CD8+T cells in spleen of WT C57BL/6 mice and HO hOX40/hOX40L/hICOS/hICOSL(2) mice by FACS. hOX40 was exclusively detectable in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice but not in wild-type mice. hOX40 was exclusively detectable on Treg, CD4+T and CD8+T cells in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice, whereas mOX40 was exclusively detectable on CD4+T cells and Treg but not on CD8+T cells in wild-type C57BL/6 mice. Leukocytes were collected from spleen of wild-type C57BL/6 mice (female, 8 weeks old) and homozygous hOX40/hOX40L/hICOS/hICOSL(2) humanized mice (female, 8 weeks old) stimulated with anti-mCD3e (50μg) in vivo for 48h, and analyzed by flow cytometry with anti-mOX40 and anti-hOX40.
Abbr. HO, homozygous; WT, wild type.

Fig.2 Detection of OX40L expression on B cells, Macrophages and DCs in spleen of WT C57BL/6 mice and HO hOX40/hOX40L/hICOS/hICOSL(2) mice by FACS. hOX40L was exclusively detectable on DCs in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice, whereas mOX40L was exclusively detectable on DCs in wild-type C57BL/6 mice. mOX40L and hOX40L expression was not detected on B cells and Macrophages in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice and wild-type mice. Leukocytes were collected from spleen of wild-type C57BL/6 mice (female, 8 weeks old) and homozygous hOX40/hOX40L/hICOS/hICOSL(2) humanized mice (female, 8 weeks old) stimulated with LPS (1μg/mL) in vitro for 48h, and analyzed by flow cytometry with anti-mOX40L and anti-hOX40L.
Abbr. HO, homozygous; WT, wild type.

Fig.3 Detection of ICOSL expression on B cells, Macrophages and DCs in spleen of WT C57BL/6 mice and HO hOX40/hOX40L/hICOS/hICOSL(2) mice by FACS. hICOSL was exclusively detectable in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice but not in wild-type mice. hICOSL was exclusively detectable on B cells, Macrophages and DCs in hOX40/hOX40L/hICOS/hICOSL(2) humanized mice, whereas mOX40 was exclusively detectable on B cells, Macrophages and DCs in wild-type C57BL/6 mice. Leukocytes were collected from spleen of wild-type C57BL/6 mice (female, 8 weeks old) and homozygous hOX40/hOX40L/hICOS/hICOSL(2) humanized mice (female, 8 weeks old) stimulated with LPS (1μg/mL) in vitro for 48h, and analyzed by flow cytometry with anti-mICOSL and anti-hICOSL.
Abbr. HO, homozygous; WT, wild type.
