Fig.1 Detection of MYOC expression in bladder, liver and cortex by RT-PCR. Human MYOC mRNA (341 bp) was detectable only in homozygous R26-CAG-MYOC mutant-3xFlag-linker-3xHA mice but not in wild-type miceBladder, liver and cortex RNA was extracted from 6-week-old male and female wild-type C57BL/6 (WT)(n=2) and homozygous R26-CAG-MYOC mutant-3xFlag-linker-3xHA mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Flag was exclusively detectable in liver and eyeball from HO R26-CAG-MYOC mutant-3xFlag-linker-3xHA mice. The liver and eyeball tissue lysates were collected from 6-week-old wild-type C57BL/6 mice and 6-week-old homozygous R26-CAG-MYOCmutant-3xFlag-linker-3xHA mice, and then analyzed by western blot with anti-Flag antibody .
