Model Description
Validation Data

Fig.1 Detection of mouse Dsg2 expression in lung and colon by RT-PCR. Mouse Dsg2 mRNA (314 bp) was detectable only in wild-type C57BL/6 mice. Total RNA of lung and colon tissues was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=3) and homozygous hDSG2 knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of human DSG2 expression in heart, lung, brain, stomach, colon and pancreas by RT-PCR. Human DSG2 mRNA (101 bp) was detectable only in homozygous hDSG2 knockin mice but not in wild-type mice. Heart, lung, brain, stomach, colon and pancreas RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=3) and homozygous hDSG2 knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.
Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.3 Detection of DSG2 expression in hDSG2 mice by WB. DSG2 was detectable in lung, heart, colon, stomach, pancreas and brain from both WT C57BL/6 and homozygous hDSG2 mice, as the antibody was cross-reactived between human and mouse DSG2. The lung, heart, colon, stomach, pancreas and brain tissue lysates were collected from 6-week-old male wild-type C57BL/6 mice and 6-week-old male homozygous hDSG2 mice, and then analyzed by western blot with anti-DSG2 antibody. Arrows showed the full-length DSG2 protein and stars showed the cleaved DSG2 protein.
Abbr. HO, homozygous; WT, wild type; PC, positive control, HepG2 cells.
