Gene Summary

Gene Symbol
Mmp9

Model Description

The endogenous mouse Mmp9 gene was replaced by human MMP9 gene.

Validation Data

Fig.1 Detection of MMP9 expression in spleen by RT-PCR. Mouse Mmp9 mRNA (548 bp) was detectable only in wild-type C57BL/6 mice. Human MMP9 mRNA (317 bp) was detectable only in homozygous hMMP9 knockin mice but not in wild-type mice. Spleen RNA was extracted from 6-week-old male wild-type C57BL/6 (WT)(n=3) and homozygous hMMP9 knockin mice (HO) (n=3), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of human MMP9(A) and mouse MMP9(B) expression in serum of WT C57BL/6 mice and HO hMMP9 mice by ELISA. Human MMP9 was exclusively detectable in homozygous hMMP9 knockin mice. Mouse MMP9 was exclusively detectable in wild-type C57BL/6 mice. Serum were collected from wild-type C57BL/6 mice (n=3, male, 6 weeks old) and HO hMMP9 mice (n=3, male, 6 weeks old), and analyzed by ELISA with anti-mMMP9 ELISA kit and anti-hMMP9 ELISA kit.

Abbr.  HO, homozygous; WT, wild type; N.D. not detected.
