Model Description

The endogenous mouse Acvr2a gene was replaced by human ACVR2A gene.

Validation Data

Fig.1 Detection of ACVR2A expression in gastrocnemius, eWAT and BAT by RT-PCR. As the mouse model was generated by mutating the mouse Acvr2a coding sequences, mRNA (411 bp) was detectable in both wild-type C57BL/6 mice and homozygous hACVR2A knockin mice using a pair of mouse Acvr2a-specific mRNA primers. Gastrocnemius, eWAT and BAT RNA were extracted from 7-week-old male wild-type C57BL/6 (WT)(n=2) and homozygous hACVR2A knockin mice (HO) (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type; eWAT, epididymal white adipose tissue; BAT, brown adipose tissue.

Fig.2 Detection of ACVR2A expression in gastrocnemius, eWAT and BAT by qPCR. Acvr2a mRNA was detectable in both wild-type C57BL/6 mice and homozygous hACVR2A knockin mice using a pair of mouse Acvr2a-specific mRNA primers, with a trend toward increased Acvr2a mRNA expression in eWAT and BAT from homozygous hACVR2A knockin mice. Gastrocnemius, eWAT and BAT RNA was extracted from 7-week-old male WT C57BL/6 and homozygous hACVR2A knockin mice (n=2 in each group), then cDNA libraries were synthesized by reverse transcription, followed by qPCR with mouse Acvr2a and mouse Gapdh primers. Relative expression represents the Acvr2a mRNA levels relative to its average expression in WT C57BL/6 mice.

Abbr. M, marker; HO, homozygous; WT, wild type; eWAT, epididymal white adipose tissue; BAT, brown adipose tissue.

Fig.3 Detection of ACVR2A expression in hACVR2A mice by WB. The skeletal muscle and eWAT tissue lysates were collected from 7-week-old male wild-type C57BL/6 mice and 7-week-old male homozygous hACVR2A mice, and then analyzed by western blot with anti-ACVR2A antibodies.

Abbr. HO, homozygous; WT, wild type. PC, positive control, Hela cells; M, marker; eWAT, epididymal white adipose tissue.

Fig.4 Detection of hACVR2A expression on mITGA7+mCD34+ muscle satellite cells in muscle of WT C57BL/6 mice and  hACVR2A knockin mice by FACS. hACVR2A positive muscle satellite cells were obviously detected with the client’s antibody. Muscle cells were collected from wild-type C57BL/6 mice (female, 8 weeks old) and hACVR2A humanized mice (female, 8 weeks old), and analyzed by flow cytometry.

Abbr. HO, homozygous; WT, wild type.
