Model Description

The endogenous mouse Npr1 gene was replaced by human NPR1 gene.

Validation Data

Fig.1 Detection of NPR1 expression in adrenal, lung and ovary by RT-PCR. Adrenal, lung and ovary RNA was extracted from 6-week-old female WT C57BL/6 (n=2) and HO hNPR1 knockin mice (n=2), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers. Mouse Npr1 mRNA (234 bp) was detectable only in wild type (WT) C57BL/6 mice. Human NPR1 mRNA (166 bp) was detectable only in homozygous (HO) hNPR1 knockin mice but not in wild-type mice.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of NPR1 expression in hNPR1 mice by WB. The heart and kidney tissue lysates were collected from 6-week-old female WT C57BL/6 mice and 6-week-old female HO hNPR1 mice, and then analyzed by western blot with anti-NPR1 antibody. NPR1 was detectable in heart and kidney from both WT C57BL/6 and HO hNPR1 mice , as the antibody was cross-reactived between human and mouse NPR1.

Abbr. HO, homozygous; WT, wild type; PC, positive control; M, marker.
