Validation Data

Fig.1 Detection of MECP2 expression in cerebellum, frontal lobe and cortex by RT-PCR. Mouse Mecp2 mRNA (488 bp) was detectable only in wild-type C57BL/6 mice. Human MECP2 mRNA (246 bp) was detectable only in homozygous hMECP2 knockin mice but not in wild-type mice. Cerebellum, frontal lobe and cortex RNA was extracted from 7-week-old male wild-type C57BL/6 (n=2) and 7-week-old male/female homozygous hMECP2 knockin mice (n=2 for each gender), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mRNA primers.

Abbr. M, marker; HO, homozygous; WT, wild type.

Fig.2 Detection of MECP2 expression in hMECP2 mice by WB. The heart, lung, cerebellum, hippocampus and cortex tissue lysates were collected from 7-week-old male wild-type C57BL/6 mice and 7-week-old male/female homozygous hMECP2 knockin mice, and then analyzed by western blot.

Abbr. HO, homozygous; WT, wild type; PC, positive control, Hela cells; M, marker.
